What is the cause of aging? Professor David Sinclair says that aging is caused by a loss in information within our epigenome. This system contains proteins such as histones, and chemical markers, like methylation, which turn genes on and off. The epigenetics allows different cell types perform their specific functions – they are what distinguishes a brain from a cell. By cosmic rays (rays of light), UV radiation (free radicals), x-rays, and cell division, our DNA is continually damaged. The epigenome does not get completely reset when our cells repair the damage. Over time, our epigenome becomes noisier. Consequently, our cells are no longer performing their functions properly.

Stressing the body can help counteract this decline. Exercise intensely, consume less protein, and experience cold. When the body detects existential threat, it activates longevity genes that attempt to maintain its body until the good times return. It may be an evolutionary legacy from early bacteria that established the two modes of life (repair, protect and grow vs. reproduce and multiply). Scientists have discovered ways to simulate stress on the body, without the discomfort that comes with fasting. The sirtuins are also activated by molecules like NMN to monitor and repair epigenome. This could slow down aging.

Yamanaka factors may help reverse aging by resetting epigenetics. These four factors are able to convert an adult cell back into a pluripotent cell. Prof. Sinclair reversed aging by using three of the factors. He discovered that they were able to see again following the treatment.
